In vitro Metabolism
Our contract service assesses in vitro metabolism studies with cryopreserved hepatocytes from various species, in suspension under continuous shaking or plated and measured at multiple time-points.
- Incubations of cryopreserved hepatocytes
- for short-term in suspension under continuous shaking
- plated, and incubated for several days, up to 7 days (learn more about long-term cultures on Pubmed)
- measuring at multiple time-points
- either the stability of the compound
- or metabolite formation
- Incubations in parallel of marker substrates as positive controls to check for metabolic competency of the test system (positive controls)
- LC/MS method development for analysis of parent and metabolites
- Full data summary
We provide:
- Highly reproducible, accurate data – validated and used by pharmaceutical, biotechnology, agrochemical and cosmetics companies, and academic organizations.
- Delivery of data within several weeks at reception of compounds, to fit in with the make-test timelines in drug discovery. A number of different reporting options are available.
- Attention to good quality customer care, with highly trained Scientists on hand to explain results and suggest the most appropriate experimental strategy.
- Flexibility – studies can be tailored to our customers’ specific requirements.
- Regulations – our services maintain and comply with regulatory guidelines providing constant confidence in the data.
Metabolic Stability
Metabolic stability influences both oral bioavailability and plasma half-life of a compound, which in turn, affect its efficacy.
KaLy-Cell evaluates the metabolic stability of xenobiotics and drug candidates in hepatocytes from various species. This analysis compares metabolic stability profiles from human and animal species in order to identify the most relevant animal model for pharmacokinetic and/or toxicity studies.
- Incubations of cryopreserved hepatocytes (e.g. human, monkey, dog, minipig, rat and mouse)
- for short-term in suspension under continuous shaking
- plated, and incubated for several days, up to 7 days, in a sandwich culture configuration allowing long-term culture and incubation
- measuring at multiple time-points the concentration of parent compound
- Incubations in parallel of marker substrates as positive controls to check for metabolic competency of the test system (positive controls)
- LC/MS method development for analysis of parent
- Full data summary
